When Steve Rogers (Chris Evans) is rejected by the army he doesn't let that stop him from trying to do his part for the war effort. During his fifth attempt he was offered a chance despite his frailness and medical conditions and grabbed onto it with both hands.
This film takes place in the Avengers film universe, and other than a reference to visits to Earth by Asgardians in the film "Thor" is the earliest indication to a divergence from the real world. To my knowledge there hasn't been a serious research project to create super soldiers by using radiation and drugs, especially not during WWII. And a great few of the weapons alongside the 'cube' technology have not been made.
However, this film wasn't made with real historical accuracy in mind.
I haven't kept up with all the characters of the Marvel universe, but from what I do know the story follows the original with a few differences. The costume more closely resembles the one from The Ultimates (Earth-1610) than the original.

If Captain America's shield represents the full amount of Vibranium in known existence, why is it wasted on a prototype which isn't even recommended?
I recommend this film if you enjoy alternate versions of WWII, and explosions aplenty.
